Hybridoma clones have been derived from hybridization of Sp2/0 myeloma cells with spleen cells of Balb/c mice immunized with synthetic N-terminal pro brain natriuretic peptide (NT-proBNP), corresponding to amino acid residues 1-12 (MAbs 5B6, 29D12), a.a.r. 13-27 (MAbs 15F11, 13G12, 18H5, 7B5, 16F3), a.a.r. 28-45 (MAbs 11D1, 16E6), a.a.r. 46-60 (MAb 15D7) and a.a.r. 61-76 (MAbs 24E11, 28F8, 15C4). The peptides used for immunization were conjugated with carrier protein.
Human NT-proBNP and proBNP.
IgG1 for MAbs 11D1, 5B6, 15D7, 7B5, 16E6, 18H5, 16F3
NT-proBNP and proBNP immunoassay. All MAbs are working in Western Blotting.
Best pairs for sandwich immunoassay (capture - detection): 15F11 - 24E11, 15C4 - 29D12, 15C4 - 13G12, 15C4 - 18H5. These pairs recognize the antigen in blood of patients with heart failure, unstable angina and myocardial infarction.
MAbs specific to peptides 5-12 (MAb 29D12) and 13-27 (MAbs 15F11, 13G12, 18H5, 7B5, 16F3) recognize circulating proBNP and can be used for the development of quantitative proBNP assays in pairs with anti-BNP MAbs (Cat.# 4BNP2). The best pairs (coating - conjugate):
