Hybridoma clones have been derived from hybridization of X63-Ag8-653 myeloma cells with spleen cells of Balb/c mice immunized with purified human proinsulin.
MAbs 3A1 and 1G4 are directed to different antigenic determinants. MAb 3A1 reacts with the site of connection of C-peptide and C-end of B-chain of proinsulin molecule.
MAb 1D6 reacts with an epitope on human proinsulin molecule, which is generated by the interaction between insulin and C-peptide parts of prohormone.
MAbs 3A1, 1G4 and 1D6 do not cross-react with human, bovine and porcine insulin, bovine and porcine proinsulin, and human C-peptide.
