TnI and TnC purified from human heart tissue.
Blood sample from the tissue donors was tested and found negative for HBsAg, HIV-1 and HIV-2 antibodies, HCV, and syphilis.
Tn I-C complex was formed by combining human cTnI and human cTnC in a molar ratio 1:2. Complex completeness was controlled in sandwich fluoroimmunoassay using an antibody pair specific for Troponin I-C complex only and an antibody pair equally reacting with free TnI and TnI in complex with TnC.
Solution in 50 mM Tris-HCl, pH 7.5, 0.15 M KCl, 5mM CaCl₂, 15 mM β-mercapthoethanol.
